Definition of Endospore Staining Endospore staining is a unique staining method, which discriminates between the two major groups of bacteria (spore formers and non-spore formers) by selectively staining the endospore against the vegetative cell. Clostridium perfringes, Bacillus cereus, Sporosarcina sp etc., are examples of endospore formers.

This complex developmental process is often initiated in response to nutrient deprivation. It allows the bacterium to produce a dormant and highly resistant cell to preserve the cell's genetic material in times of extreme stress. Endospores can survive environmental assaults that would normally kill the bacterium.

The main purpose of endospore staining is to differentiate bacterial spores from other vegetative cells and to differentiate spore formers from non-spore formers. When vegetative cells of certain bacteria such as Bacillus spp and Clostridium spp are subjected to environmental stresses such as nutrient deprivation, they produce metabolically inactive or dormant form-endospore. Formation of endospores circumvent the problems associated with environmental stress and helps them to survive.
